As an endocrine organ, the pancreatic islet secretes several hormones including insulin and glucagon. However, to date no studies have focused on the pancreatic islet secretome. Therefore, it is necessary to profile the pancreatic islet secretome, which may be dependent on the glucose condition, and to identify changes in the secreted protein pool following various glucose concentration stimuli. In this study, primary islets were prepared from C57BL/6 adult mice and treated with various glucose concentrations. A comparative secretomic analysis was then conducted. Ions and glucose were removed by a centrifugal filter in conditioned media and fractionated. The exosomes were then analyzed for peptides by liquid chromatography-electrospray ionization mass spectrometry (LC-ESI-MS/MS). Proteins such as MIF, ENO2, and GSTM1 were identified after filtering. We found an increase in ENO2 and MIF and a decrease in GSTM1 in the high glucose condition compared to the low glucose condition. This was the first systematic unbiased study of the pancreatic islet secretome, and the results suggest that novel secretory factors may be dependent on glucose levels.
